Her defiant ignorance makes it a joke about ignoring rules, warnings and instructions, either because you didn't read them or because you've decided they don't apply to you. The sign is blank so you can write any rule on it.
Origin: The scene is from the PBS cartoon Arthur, in a late-1990s episode featuring D.W. Fans later blanked the sign so it could be relabeled with any warning.
